为阐明黄鳝大型多钩槽绦虫渝西地区分离株线粒体基因细胞色素c氧化酶第I亚基(cox1)的遗传变异情况,本试验对黄鳝大型多钩槽绦虫cox1序列进行PCR扩增、克隆、测序及序列分析,并与GenBank中已知绦虫相应基因序列进行比较分析.结果显示渝西地区6个样品cox1碱基序列长度一致,均为444bp.同源分析发现渝西地区不同分离株间的同源性较高,为97.3%~100%,而6个大型多钩槽绦虫分离株与GenBank中收录的其他绦虫相比,同源性较低,为10.1%~12.1%.由于大型多钩槽绦虫cox1序列种内相对保守,种间差异较大,故可以作为种间遗传变异研究的标记.
In order to elucidate the genetic variation of mitochondrial cytochrome c oxidase I subunit (cox1) in the large-scale Echinococcus multilocularis in western Chongqing, Sequencing and sequence analysis, and compared with the corresponding gene sequences of the known tapeworm in GenBank.The results showed that the cox1 base sequences of the 6 samples in the West of Chongqing were the same in length, both of which were 444bp.The homology analysis showed that among the isolates The homology was high from 97.3% to 100%, while the isolates from 6 large polygalactosides had low homology of 10.1% ~ 12.1% compared with other tapeworms in GenBank. Coxsackie cox1 sequences within the relatively conservative species, species differences between larger, it can be used as a marker of genetic variation among species.
