目的:观察生地大黄汤对脑出血(ICH)大鼠的神经保护作用。方法:将60只大鼠随机分为假手术组、ICH模型组、生地大黄汤组。假手术组只开颅钻孔进针,后两组采用自体血注入法制备大鼠脑出血模型。于造模后6 h,生地大黄汤组灌胃生地大黄汤,假手术组和ICH模型组灌胃等量生理盐水。分别于造模后3 h、1 d、3 d进行神经功能评分;造模后1 d、3 d测定脑含水量及观察脑组织病理学。结果:造模后3 h与1 d,生地大黄汤组神经功能缺损评分与ICH模型组比较均无统计学差异(P>0.05);造模后3 d,生地大黄汤组神经功能缺损评分明显低于ICH模型组(P<0.05)。ICH模型组与生地大黄汤组随着出血时间延长脑含水量均呈逐渐增高趋势,但在造模后3 d时生地大黄汤组脑含水量较ICH模型组明显减少(P<0.01)。造模后3 d,ICH模型组血肿周围区域神经元胞体明显变小,尼氏体明显减少;与假手术组比较,生地大黄汤组、ICH模型组尼氏体在各时间点的计数皆减少(P<0.01),而造模后3 d时生地大黄汤组尼氏体计数明显高于ICH模型组(P<0.01)。结论:生地大黄汤对脑出血大鼠有确切的神经保护作用,能降低神经功能缺损评分,减轻脑水肿程度,增加神经细胞功能活性,有效促进脑出血损伤恢复。
Objective: To observe the neuroprotective effect of Shengdi Dahuang Decoction on cerebral hemorrhage (ICH) rats. Methods: Sixty rats were randomly divided into sham-operation group, ICH model group and Sheng-Di-Da-Huang Decoction group. Sham operation group only craniotomy drilling needle, the latter two groups using autologous blood injection rat cerebral hemorrhage model. After 6 h of modeling, Dahuang Decoction group was fed with rhubarb decoction, and the sham operation group and ICH model group were given the same amount of normal saline. Neurological function scores were made at 3 h, 1 d, and 3 d after modeling, respectively. Brain water content and brain histopathology were measured at 1 and 3 days after modeling. Results: After 3 h and 1 d of modeling, the neurological deficit score of Shengdai Dazhuang Decoction group was not significantly different from that of ICH model group (P> 0.05) Lower than ICH model group (P <0.05). Compared with ICH model group, ICH model group and Shengdai Dhuang Decoction group showed a gradually increasing trend of brain water content with prolonged bleeding time. However, the brain water content of ICH model group and ICD model group decreased significantly (P <0.01) 3 days after modeling. At 3 days after model establishment, the neuronal soma in the area around the hematoma in ICH model group was significantly smaller and the Nissl body was significantly reduced. Compared with the sham operation group, the counts of Nissl body in the ICH model group were decreased at different time points (P <0.01). The number of Nissl body of Shengdi Dahuang Decoction group was significantly higher than that of ICH model group (P <0.01) on the 3rd day after model establishment. Conclusion: Radix Rehmanniae Decoction has the exact neuroprotective effect on rats with intracerebral hemorrhage, which can reduce the score of neurological deficit, reduce the degree of cerebral edema, increase the functional activity of nerve cells and effectively promote the recovery of cerebral hemorrhage.
