目的:探讨不同剂量的阿托伐他汀对大鼠局灶性脑缺血后胡须体觉皮层功能的影响及其机制。方法:30只大鼠训练达标后依随机数字表法分为假手术组、对照组(0.1ml/kg生理盐水)、低剂量组(1.0mg/kg)、中剂量组(3.0mg/kg)、高剂量组(6.0mg/kg)。对照组及各剂量组采用显微镜下结扎大脑中动脉数根分支构建局灶性胡须体觉皮层缺血模型并分别于造模后24h开始灌胃,每天1次,连续7天。造模后每天进行胡须依赖实验检测直至达到标准并于术后14天,应用超声多普勒检测缺血区周围的血流量;应用免疫组化检测缺血区周围CD34及VEGF的表达情况。结果:与对照组相比,低、中、高剂量组均增强了胡须依赖辨别能力(P<0.01,P<0.01,P<0.05);同时提高了缺血区周围的血流量(P<0.01,P<0.01,P<0.05);增加了缺血区周围CD34、VEGF阳性细胞数(P<0.01,P<0.01,P<0.05)。而且中剂量组与低剂量、高剂量组相比,差异有统计学意义(P<0.05,P<0.01)。结论:阿托伐他汀有助于缺血后胡须依赖辨别能力的改善,其机制可能通过促进VEGF的表达及增加局部的微血管密度从而提高局部血流,且以中剂量效果较好。
Objective: To investigate the effects of different doses of atorvastatin on bearded somatic cortex function after focal cerebral ischemia in rats and its mechanism. Methods: Thirty rats were randomly divided into sham-operation group, control group (0.1ml / kg normal saline), low dose group (1.0mg / kg) and middle dose group (3.0mg / kg) , High dose group (6.0mg / kg). The control group and each dose group were given focal ligation of the middle cerebral artery branches under the microscope to establish the focal beard somatosensory cortex ischemia model and were intragastrically administered intragastrically once a day for 24 hours. The beard-dependent test was performed daily until the standard was reached and the blood flow around the ischemic area was detected by Doppler ultrasound 14 days after the operation. The expression of CD34 and VEGF around the ischemic area was detected by immunohistochemistry. Results: Compared with the control group, the beard-dependent discrimination ability was enhanced (P <0.01, P <0.01, P <0.05), and the blood flow around the ischemic zone was increased , P <0.01, P <0.05). The number of CD34 and VEGF positive cells around the ischemic area increased (P <0.01, P <0.01, P <0.05). And the difference between the middle dose group and the low dose and high dose group was statistically significant (P <0.05, P <0.01). Conclusion: Atorvastatin can improve the beard-dependent discrimination ability after ischemia, and its mechanism may improve local blood flow by promoting the expression of VEGF and increasing local microvessel density, and the effect of middle dose is better.
