目的探讨皮质抑素(cortistatin,CST)在人肾组织中的表达情况及其在不同分级IgA肾病肾组织中的表达变化。方法收集10例肾癌切除术患者的癌旁正常肾组织,通过逆转录聚合酶链反应(RT-PCR)与Western印迹分别检测人肾组织中CST mRrNA与蛋白的表达情况,采用免疫组织化学(IHC)技术对CST于肾组织中的表达进行定位。根据Lee分级标准将IgA肾病分为3组:Ⅰ~Ⅱ级为A组、Ⅲ~Ⅳ级为B组、Ⅴ级为C组,每组分别收集10例IgA肾病患者的肾穿组织样本。采用IHC技术,检测正常肾组织及不同分级IgA肾病肾组织中CST的表达变化。对临床指标进行多重线性回归分析,评估其对IgA肾病肾组织内CST表达量的影响。结果 RT-PCR与Western印迹结果均表明肾组织内表达CST,IHC结果显示CST表达于肾小管上皮细胞。IgA肾病中,病理分级越高,肾小管中CST表达越多。多重线性回归分析显示,病理分级与肾组织中CST表达量相关(r=0.875,尸<0.01)。结论 CST可能参与IgA肾病病理损伤的炎症反应,发挥抗炎保护作用。
Objective To investigate the expression of cortistatin (CST) in human renal tissues and its changes in different grade IgA nephropathies. Methods Ten normal renal tissues adjacent to the renal cell carcinoma were collected. The expression of CGRR and protein in human renal tissues were detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot, respectively. Immunohistochemistry IHC) technology to locate the expression of CST in renal tissues. IgA nephropathy was divided into 3 groups according to Lee classification: group Ⅰ ~ Ⅱ was group A, group Ⅲ ~ Ⅳ was group B, group V was group C, and renal interstitial samples of 10 patients with IgA nephropathy were collected in each group. IHC technique was used to detect the expression of CST in normal kidney tissues and renal tissues with different grade IgA nephropathy. Multiple linear regression analysis of clinical indicators to assess its impact on renal CST expression in IgA nephropathy. Results RT-PCR and Western blotting results showed that CST was expressed in renal tissues, and IHC results showed that CST was expressed in renal tubular epithelial cells. IgA nephropathy, the higher the pathological grade, the more tubular CST expression. Multiple linear regression analysis showed that the pathological grade correlated with the expression of CST in renal tissues (r = 0.875, P <0.01). Conclusion CST may be involved in the inflammatory reaction of pathological injury of IgA nephropathy and exert anti-inflammatory protection.
