目的 研究克拉霉素对小鼠 L ewis肺癌的治疗效果和作用机制 ,为临床应用提供实验依据 .方法 C5 7BL/ 6小鼠右肋处皮下接种 L ewis肺癌 ,6 h后随机分为 2组 ,每组 15只 ,分别接受克拉霉素 5 0 mg· kg- 1· d- 1 (实验组 )或等体积 5 %葡萄糖 (对照组 )腹腔内注射 ,每日 1次 ,连用 2 1d.观察比较实验组与对照组小鼠存活时间、移植瘤体积、肿瘤组织内微血管密度和肺脏转移瘤数目 .结果 实验组小鼠平均存活时间为 (2 7.0± 2 .6 ) d,肿瘤组织内微血管密度为 18.7± 11.4,肺脏表面的转移瘤数目为12 .0± 2 .9,对照组小鼠的上述参数分别为 (19.1± 2 .0 ) d,2 4.5± 13.7和 18.6± 5 .4,两组差异非常显著 (P<0 .0 5 ) .结论 克拉霉素具有抗肿瘤血管生成和抗肿瘤转移的作用 ,在肿瘤的综合治疗中具有一定的应用价值 .
Objective To study the therapeutic effect and mechanism of clarithromycin on L ewis lung cancer in mice and provide experimental basis for its clinical application.Methods L7 lung cancer was inoculated subcutaneously in the right rib of C5 7BL / 6 mice and then randomly divided into 2 groups after 6 h, Each group received intraperitoneal injection of clarithromycin 50 mg · kg-1 · d-1 (experimental group) or equal volume of 5% glucose (control group) intraperitoneally once daily for 21 days. The survival time, the volume of tumor xenografts, the microvessel density and the number of lung metastases in the experimental group and the control group were observed.Results The average survival time of the mice in the experimental group was (2 7.0 ± 2.66) d, the microvessel density in the tumor tissue was 18.7 ± 11.4, the number of metastases on the surface of the lung was 12.0 ± 2.9, the above parameters in the control group were (19.1 ± 2.0) days, 4.5 ± 13.7 and 18.6 ± 5.4, respectively (P <0.05) .Conclusion Clarithromycin has antitumor angiogenesis and anti-tumor metastasis and has certain value in the comprehensive treatment of tumors.
