目的:建立测定牛蒡草主要活性成分绿原酸含量的方法,并比较不同产地牛蒡草绿原酸的含量差异,为牛蒡草质量控制提供依据。方法:采用Agilent C18色谱柱(4.6 mm×150 mm,5μm),以乙腈-0.4%磷酸水溶液(8∶92)为流动相,检测波长327 nm,体积流量1.0 mL·min-1,柱温30℃,进样量10μL,外标法定量。结果:绿原酸得到良好分离,在0.041 8~4.18μg与峰面积积分值呈良好的线性关系,平均加样回收率为100.3%。结论:该方法简便、准确、分离度良好,为牛蒡草的质量控制提供了依据。
OBJECTIVE: To establish a method for the determination of chlorogenic acid in the main active ingredient of Arctium lappa L. and to compare the differences in the contents of chlorogenic acid in Arctium lappa L. in different areas so as to provide the basis for the quality control of Arctium lappa. Methods: Agilent C18 column (4.6 mm × 150 mm, 5 μm) was used with mobile phase of acetonitrile-0.4% phosphoric acid (8:92) as mobile phase at a detection wavelength of 327 nm with a volume flow rate of 1.0 mL · min- ℃, injection volume 10μL, external standard method. Results: Chlorogenic acid was well separated. The linear range was 0.041 8 ~ 4.18μg with the peak area integral value, and the average recovery was 100.3%. Conclusion: The method is simple, accurate and well-separated and provides the basis for the quality control of Arctium lappa.
