目的研究负载肿瘤抗原的DC疫苗对T739小鼠体内膀胱肿瘤的免疫学效应。方法建立BTT739荷瘤小鼠动物模型,随机分为实验组和实验对照组和空白对照组,于肿瘤细胞接种后第7、14天给予相应DC疫苗治疗,每组分2亚组,分别用于测量瘤质量、体积及用于观察荷瘤小鼠存活情况。结果负载肿瘤抗原的DC疫苗实验组荷瘤鼠肿瘤平均体积和平均瘤质量均显著低于对照组(P<0.01),生存期长于对照组,并且有2只小鼠无瘤长期存活。经DC疫苗实验组治愈的2只小鼠30d后无肿瘤生长,再次皮下接种BTT739细胞观察30d仍无肿瘤发生。结论负载肿瘤抗原的DC疫苗对膀胱肿瘤荷瘤小鼠具有抑瘤效应和生存期延长作用。
Objective To study the immunological effect of DC vaccine loaded with tumor antigen on bladder tumor in T739 mice. Methods The animal model of BTT739 tumor-bearing mice was established and randomly divided into experimental group, experimental control group and blank control group. Corresponding DC vaccines were given on the 7th and 14th day after inoculation of tumor cells, and each subgroup was divided into 2 subgroups, Measurement of tumor mass, volume and used to observe the survival of tumor-bearing mice. Results The average tumor volume and the average tumor mass of the DC vaccine bearing tumor antigen in the experimental group were significantly lower than those in the control group (P <0.01), and the survival time was longer than that in the control group. Two mice survived without tumor. Two mice cured by DC vaccine experimental group had no tumor growth after 30 days, and again there was no tumorigenesis after subcutaneous inoculation with BTT739 cells for 30 days. Conclusion The DC vaccine loaded with tumor antigen has antitumor effect and prolongation of survival in bladder tumor bearing mice.
