目的研究SBA-15对人脐静脉血管内皮细胞(human umbilical vein endothelial cells,HUVEC)的细胞活性和炎症因子C-反应蛋白(CRP)、单核细胞趋化蛋白-1(MCP-1)mRNA表达水平的影响。方法分别将HUVEC细胞暴露于含有终浓度为0(溶剂对照)、25、50、100、200μg/ml的SBA-15和100μg/ml微米二氧化硅(阳性对照)RPMI1640培养基中培养24 h。测定HUVEC增殖的抑制情况、培养液上清中乳酸脱氢酶(LDH)的活力及CRP和MCP-1 mRNA的表达水平。结果与溶剂对照组相比,SBA-15染毒HUVEC的抑制率、培养液上清中LDH活力和CRP和MCP-1 mRNA的表达水平均显著升高;且随着SBA-15染毒浓度的升高,HUVEC的抑制率、培养液上清中LDH活力和CRP和MCP-1 mRNA的表达水平均呈上升趋势。结论 SBA-15对HUVEC具有毒性作用。
Objective To investigate the effects of SBA-15 on the cell viability and the expressions of MCP-1 mRNA and cytokines in human umbilical vein endothelial cells (HUVEC) Horizontal impact. Methods HUVEC cells were respectively cultured for 24 h in RPMI 1640 medium containing SBA-15 at 25, 50, 100, 200 μg / ml and 100 μg / ml silica (positive control) at a final concentration of 0 (solvent control). The inhibition of proliferation of HUVEC, the activity of lactate dehydrogenase (LDH) and the expression of CRP and MCP-1 mRNA in the culture supernatant were measured. Results Compared with the solvent control group, the inhibitory rate of HUVEC induced by SBA-15 and the expression of LDH and CRP and MCP-1 mRNA in supernatant of culture medium were significantly increased. With the increase of SBA-15 concentration The inhibitory rate of HUVEC, the activity of LDH and the expressions of CRP and MCP-1 mRNA in culture supernatant increased. Conclusion SBA-15 has a toxic effect on HUVEC.
