目的:建立检测芦笋中毒死蜱残留的ELISA方法。方法:用间接竞争ELISA法建立标准抑制曲线并测定交叉反应率、回收率,并与GC结果对比。结果:毒死蜱与其特异性单克隆抗体竞争性结合的半数抑制浓度(Median inhibitoryconcentration,IC50)为1.45μg/ml,该检测法的检出限为0.027μg/ml。芦笋样品经简单前处理后分别采用ELISA和GC测定,用ELISA方法测得芦笋中毒死蜱的添加回收率为82.60%~98.96%,与GC测定结果基本相当。结论:成功建立了检测芦笋中毒死蜱残留的ELISA方法,为今后更深入的ELISA研究奠定了基础。
Objective: To establish an ELISA method for the determination of chlorpyrifos residues in asparagus. Methods: The standard inhibition curve was established by indirect competitive ELISA and the cross-reactivity and recovery rates were determined. The results were compared with the GC results. Results: The IC50 of competitive inhibition of chlorpyrifos and its specific monoclonal antibody was 1.45μg / ml. The detection limit of this assay was 0.027μg / ml. Asparagus samples were detected by ELISA and GC respectively after simple pretreatment. The recoveries of chlorpyrifos in the asparagus were 82.60% ~ 98.96% by ELISA, which were basically the same as the results of GC. Conclusion: The ELISA method for the determination of chlorpyrifos residues in asparagus has been successfully established, which lays the foundation for further ELISA research in the future.
