目的寻找一种快速简便的蒲公英干品总DNA的提取方法。方法用改良CTAB、SDS、高盐低pH 3种提取方法及液氮、玻璃砂2种研磨方法提取蒲公英总DNA,采用蛋白核酸测定及电泳检测所得DNA的纯度和浓度。结果 3种提取方法所得到的DNA纯度和浓度不同,改良CTAB法提取总DNA纯度和浓度较高,电泳显示主条带较清晰,降解较少;2种研磨方法蛋白核酸测定及电泳结果相近。结论改良CTAB法适用于蒲公英药材基因组DNA的提取。
Objective To find a rapid and simple method for extracting total DNA from dried dandelion stem. Methods The total DNA of dandelion was extracted by three extraction methods: improved CTAB, SDS, high salt and low pH, and two kinds of grinding methods of liquid nitrogen and glass sand. The purity and concentration of DNA were determined by protein nucleic acid assay and electrophoresis. Results The purity and concentration of DNA extracted by three extraction methods were different. The purity and concentration of total DNA extracted by modified CTAB method were higher. The electrophoresis showed that the main band was clearer and had less degradation. The results of two methods were similar. Conclusion The modified CTAB method is suitable for the extraction of genomic DNA of dandelion herbs.
