探讨氯卡色林(lorcaserin)对高脂饮食小鼠瘦素及胰岛素信号的影响。30只C57BL/6小鼠随机分为正常组、模型组和给药组;正常组给予标准饲料,模型及给药组给予高脂饲料;同时,给药组每天腹腔注射氯卡色林(10 mg/kg),而其余两组腹腔注射等体积生理盐水;每周测定小鼠体重,直至第12 w取材;试剂盒测定血清胆固醇、甘油三酯、高密度脂蛋白、低密度脂蛋白含量;ELISA法测定血清瘦素和胰岛素水平;q PCR技术检测下丘脑瘦素受体和胰岛素受体基因表达。结果表明,氯卡色林能够有效抑制高脂饲料饮食小鼠体重增加,降低血清胆固醇含量,提高血清胰岛素水平,同时增强下丘脑瘦素受体基因表达。实验结果提示,氯卡色林减肥作用机制与其增强胰岛素和瘦素信号传导有关。
To investigate the effects of lorcaserin on leptin and insulin signaling in high fat diet mice. Thirty C57BL / 6 mice were randomly divided into normal group, model group and administration group. In the normal group, high fat diet was given to the model group and the administration group. At the same time, the rats in the administration group were given intraperitoneal injection of lorcaserin mg / kg), while the other two groups were injected intraperitoneally with equal volume of normal saline. The body weight of mice was measured weekly until the 12th wk. The contents of serum cholesterol, triglyceride, high density lipoprotein, low density lipoprotein, Serum leptin and insulin levels were measured by ELISA. The expression of leptin receptor and insulin receptor mRNA in hypothalamus was detected by q PCR. The results showed that lorcaserin can effectively inhibit high fat diet mice weight gain, lower serum cholesterol, increase serum insulin levels, while enhancing hypothalamic leptin receptor gene expression. Experimental results suggest that lorcaserin weight loss mechanism and its enhanced insulin and leptin signaling related.
