目的 建立嘌呤 +胞嘧啶摩尔百分比 (GC % )的测定细菌染色体方法。方法 以磷酸二氢铵、乙腈混合液为流动相 ,选择逆相分配 (5C18) ,通过紫外分光检测器分析DNA样品中四种硷基的相对含量 ,并计算出GCmol%。结果 16株代表高、中、低GC %细菌的测定结果与发光值 (Tm/Bd法 )完全相符 ,其相对不精密度小于 1 0 3%。结论 此法检测的细菌GC %值精确可靠 ,可用作细菌的分类与鉴定
Objective To establish a method for the determination of bacterial chromosome by purine + cytosine molar percentage (GC%). Methods The mobile phase consisted of ammonium dihydrogen phosphate and acetonitrile as the mobile phase. Reverse phase partitioning (5C18) was selected. The relative content of four basic groups in DNA samples was analyzed by ultraviolet spectrophotometer and GCmol% was calculated. Results The results of determination of 16 high, medium and low GC% bacteria were completely consistent with the luminescence (Tm / Bd) method. The relative imprecision was less than 103%. Conclusion The GC% of bacteria detected by this method is accurate and reliable, and can be used as the classification and identification of bacteria
