目的探讨东宫泉对黑素细胞增殖及其酪氨酸酶活性的影响。方法体外培养小鼠B16黑素瘤细胞,倒置显微镜下观察不同浓度东宫泉(将50倍浓缩液0、2、4、8、16、32、64倍稀释)对B16黑素瘤细胞形态的影响,MTT法检测不同浓度东宫泉对细胞增殖的影响,蘑菇酪氨酸酶多巴速率氧化法测定不同浓度东宫泉对B16黑素瘤细胞酪氨酸酶活性的影响。结果经16倍稀释的东宫泉处理48h的B16黑素瘤细胞生长旺盛;东宫泉对B16黑素瘤细胞具有高浓度抑制、中低浓度激活的作用,8倍和16倍稀释为最佳激活浓度(P<0.05);东宫泉对酪氨酸酶亦具有高浓度抑制、低浓度激活的作用。结论东宫泉对黑素细胞增殖和酪氨酸酶均具有双向调节作用。
Objective To investigate the effect of Donggongquan on melanocyte proliferation and tyrosinase activity. Methods B16 melanoma cells were cultured in vitro. The effects of different concentrations of Donggongquan (0, 2, 4, 8, 16, 32, 64 times dilution) on B16 melanoma cells were observed under inverted microscope The effects of different concentrations of Donggongquan on the proliferation of B16 melanoma cells were detected by MTT assay. The effect of different concentrations of Donggongquan on the tyrosinase activity of B16 melanoma cells was determined by the mushroom tyrosinase Doppler rate oxidation method. Results The 16-day-old Dongguangquan 48h B16 melanoma cells grew vigorously; Donggongquan had a high concentration of B16 melanoma cells inhibition, the role of medium and low concentrations of activation, 8-fold and 16-fold dilution of the best activation concentration (P <0.05). Donggongquan also has a high concentration of tyrosinase inhibition, low-concentration activation. Conclusion Donggongquan has a two-way regulation of melanocyte proliferation and tyrosinase.
