目的:研究人肾癌组织中EphB4基因及其蛋白的表达及其在肾癌发生、发展中的作用和临床意义。方法:取液氮保存的原发性肾癌新鲜标本40例,相应癌旁组织(阴性切缘)40例,正常肾脏组织30例,采用半定量逆转录PCR(RT-PCR)和免疫组化SP法检测EphB4基因及其蛋白表达水平,并结合临床病理资料进行分析。结果:EphB4基因在肾癌组织和癌旁组织中都表达,且癌旁表达低于肿瘤表达,差异有统计意义(P<0.05);正常肾脏组织不表达(P<0.01);40例肾癌标本中,25例EphB4蛋白表达阳性,阳性率为62.5%,40癌旁组织中,10例EphB4蛋白表达阳性,阳性率为25.0%;两者有显著差异性(P<0.05)。EphB4蛋白的表达与患者的性别、年龄无关(P>0.05),但与肿瘤浸润深度、肿瘤分化程度、淋巴结转移密切相关(P<0.05)。结论:原癌基因EphB4及其蛋白在人肾癌组织中过度表达,且与肾癌的发生,发展相关,可能成为肾癌预后的侯选标志物之一。
Objective: To investigate the expression of EphB4 gene and its protein in human renal cell carcinoma and its role in the occurrence and development of renal cell carcinoma and its clinical significance. Methods: Forty fresh specimens of primary renal cell carcinoma preserved by liquid nitrogen were collected. Forty corresponding cases of paracancerous tissues (negative margin) and 30 cases of normal renal tissues were obtained. The mRNA expression levels were determined by semi-quantitative RT-PCR and immunohistochemistry SP method to detect EphB4 gene and its protein expression levels, combined with clinical and pathological data analysis. Results: EphB4 gene was expressed in both renal cell carcinoma and paracancerous tissues, and the expression of EphB4 was lower than that in tumor tissues (P <0.05), but not in normal kidney tissues (P <0.01) The positive expression rate of EphB4 protein in the specimens was 62.5%. EphB4 protein expression was positive in 10 of 40 adjacent tissues, with a positive rate of 25.0%. There was significant difference between the two groups (P <0.05). The expression of EphB4 protein was not related to the gender and age of patients (P> 0.05), but closely related to the depth of tumor invasion, tumor differentiation and lymph node metastasis (P <0.05). CONCLUSION: The proto-oncogene EphB4 and its protein overexpressed in human renal cell carcinoma and related to the occurrence and development of renal cell carcinoma may be one of the candidate markers for the prognosis of renal cell carcinoma.
