Objective: To provide theoretical basis for the drug treatment of RA by studying effect of triptolide on the gene of collagen-induced arthritis rats joint synovial cells.Methods: After CIA rats synovial cell were treated with triptolide,to detect CIA rats synovial cell proliferation by MTT method.The percentage of apoptosis synovial cells were detected by Annexin V/PI.The mRNA expression of DR3、PDCD5、 FasL、 sFas、 DNMT1、DNMT3a and DNMT3b of collagen-induced arthritis rats synovial cells were detected by realtime PCR.Results: Triptolide(10.0 ηg/mL to 160.0 ηg/mL) can inhibit proliferation of the CIA rats synovial cells and presented good dose dependent.Additionally,the mRNA expression of proapoptotic genes DR3 and FasL were remarkably increased after being administered with triptolide.The mRNA expression of DNMT1 and DNMT3a were remarkably reduceded after being administered with triptolide.Conclusion: One of the molecular mechanisms that triptolide promote CIA rats synovial cells apoptosis was that it can reduce the expression of DNMT1 and DNMT3a,which could increase the expression of proapoptotic genes (DR3 and FasL) and increased their apoptosis.These findings were to provide a scientific and systematic theoretical basis for the medicinal application of triptolide and the treatment of autoimmune arthritis.
