The mechanism consists of a tumor-suppressor protein (TSP) containing a DNA-binding domain (DBD) and RNAbinding domains (RBD), and a RNA, usually a non-coding RNA (ncRNA).The DBD binds to proto-oncogenes (POG), repressing transcription of POG and suppressing tumorigenesis, and the ncRNA binds to the RBD, releasing the TSP from POG and activating transcription and tumorigenesis.Embryonic cells express high levels of TSP-binding ncRNA that drive cell proliferation by preventing repression of POG.The level of TSP-binding ncRNA subsequently decreases in most differentiating cells, enabling a TSP to repress POG and stop cell proliferation.Tumorigenesis can result when the level of TSP-binding ncRNA fails to decrease in an embryonic cell or increases in a differentiated cell.This mechanism also regulates transcription of P450scc, the first gene in the steroidogenic pathway.A TSP-binding ncRNA provides a potential diagnostic and therapeutic target for cancer.
