目的探讨成人隐匿性自身免疫性糖尿病(LADA)患者外周血细胞因子白介素-10(IL-10)、转化生长因子-β(TGF-β)水平变化及其与胰岛β细胞功能的关系。方法以双抗夹心ELISA法检测30例LADA(LADA组)、30例2型糖尿病(T2DM组)、30例正常对照(正常对照组)外周血血清中IL-10、TGF-β水平,同时应用HOMA公式计算胰岛素释放指数(HOMA-IS)。结果LADA组的IL-10水平低于T2DM组(P<0.05),高于正常对照组(P<0.05)。LADA组的TGF-β水平与T2DM组相比,差异无统计学意义(P>0.05)。IL-10水平与患者LnHOMA-IS成正相关(r=0.314,P=0.017)。结论LADA患者体内存在细胞免疫功能失衡,不能有效地维持自身的免疫耐受,可能是LADA患者胰岛β细胞功能损害的主要因素。
Objective To investigate the changes of peripheral blood cytokines interleukin-10 (IL-10) and transforming growth factor-β (TGF-β) levels in adult patients with latent autoimmune diabetes (LADA) and their relationship with the function of islet β cells. Methods Serum levels of IL-10 and TGF-β in 30 cases of LADA group, 30 cases of type 2 diabetes mellitus (T2DM group) and 30 normal controls (normal control group) were detected by double-antibody sandwich ELISA. HOMA formula to calculate the insulin release index (HOMA-IS). Results The level of IL-10 in LADA group was lower than that in T2DM group (P <0.05), higher than that in normal control group (P <0.05). TGF-βlevels in LADA group were not significantly different from those in T2DM group (P> 0.05). The level of IL-10 was positively correlated with LnHOMA-IS (r = 0.314, P = 0.017). Conclusions The imbalance of cellular immune function in LADA patients can not effectively maintain their own immune tolerance, which may be the main factor of pancreatic β-cell dysfunction in LADA patients.
