目的:从肠道吸收、肝脏摄取的角度解释戊己丸的配伍机制。方法:采用正交设计得到9个戊己丸复方,加上9个单味药,共18个复方为对象。建立单次肠肝灌流模型,于十二指肠给药后0、1、5、10、15、30、60、90、120min在门静脉、肝静脉插管处同时取样,LC-MS检测代表成分浓度,计算药-时曲线下面积(AUC)、肝脏摄取率,并对结果进行方差分析、极差分析。结果:吴茱萸与黄连代表成分小檗碱、巴马汀、白芍代表成分芍药苷吸收呈负相关,白芍与黄连代表成分小檗碱、巴马汀吸收呈正相关,而与吴茱萸代表成分吴茱萸碱、吴茱萸次碱吸收呈负相关;吴茱萸吴茱萸碱、吴茱萸次碱与黄连代表成分小檗碱、巴马汀肝脏摄取呈正相关,黄连与白芍代表成分芍药苷肝脏摄取呈负相关,白芍与黄连代表成分小檗碱、巴马汀肝脏摄取呈负相关。并且得到了针对代表成分吸收、肝脏摄取的最优配伍比例。结论:吴茱萸抑制黄连、白芍代表成分的吸收;白芍促进黄连代表成分、抑制吴茱萸代表成分的吸收。吴茱萸促进黄连代表成分的肝脏摄取;黄连、白芍相互抑制其代表成分的肝脏摄取。
Objective: To explain the compatibility mechanism of Wu-hui Wan from the perspective of intestinal absorption and liver uptake. Methods: By using orthogonal design, we obtained nine compounds of Wuji Pills, plus nine single herbs and a total of 18 compounds. A single intestinal perfused hepatic perfusion model was established. And got the proportion of the optimal compatibility for the absorption of the representative ingredient and the liver. Conclusion: Fructus Evodiae can inhibit the absorption of the representative components of Coptis Chinensis and Radix Paeoniae Alba, while Radix Paeoniae Alba is able to promote the absorption of the representative components of Fructus Evodiae. Fructus Evodiae can promote the liver uptake of the representative ingredients of Coptis chinensis; Coptis chinensis and Radix Paeoniae Alba can inhibit the liver uptake of its representative components.
