目的:研究解毒凉血方对肝卵圆细胞的诱导分化作用及其作用机制。方法:SD大鼠随机分为模型组、解毒凉血方组、正常对照组;通过喂饲2-乙酰氨基芴加2/3肝切除术建立肝卵圆细胞增殖模型;HE染色观察肝组织病理学,透射电镜观察肝卵圆细胞的超微结构,免疫组织化学观察各组大鼠肝组织OV6、ALB、CD34及Notch1的表达和分布。结果:模型组造模过程中至7周末肝组织汇管区周围可见大量的卵圆细胞增生,OV6、CD34、Notch1蛋白与正常对照组比较在肝组织中呈高表达,ALB蛋白较正常对照组表达明显减少;解毒凉血方组肝组织卵圆细胞增生较模型组明显减少,OV6、CD34、Notch1蛋白表达较模型组明显减少(P<0.05)、ALB蛋白表达较模型组明显增加(P<0.05)。结论:解毒凉血方能有效的促进卵圆细胞向成熟肝细胞分化,此作用机制可能与调节Notch信号通路有关。
Objective: To study the induction and differentiation of hepatic oval cells by Jiedu Liangxue prescription and its mechanism. Methods: SD rats were randomly divided into model group, Jiedu Liangxue Fang group and normal control group; hepatic oval cell proliferation model was established by feeding 2-acetylaminofluorene plus 2/3 hepatectomy; HE staining was used to observe liver tissue disease The ultrastructures of hepatic oval cells were observed under transmission electron microscope and transmission electron microscope. The expression and distribution of OV6, ALB, CD34 and Notch1 in liver tissues were observed by immunohistochemistry. RESULTS: A large number of oval cells proliferated around the portal area of the hepatic tissue during 7 weeks after modeling in the model group. OV6, CD34 and Notch1 proteins were highly expressed in liver tissue compared with the normal control group, while ALB protein was higher than that in the normal control group (P <0.05). The protein expression of ALB was significantly increased compared with the model group (P <0.05), and the expression of OV6, CD34 and Notch1 protein was significantly decreased ). Conclusion: Detoxification and cooling blood can effectively promote the differentiation of oval cells into mature hepatocytes. The mechanism may be related to the regulation of Notch signaling pathway.
