The pro-death Bcl-2 family protein and tumor suppressor Bax is frequently mutated in tumors with microsatellite instability (MSI).A single nucleotide deletion in the Bax exon 3 microsatellite region leads to a reading frame-shift and "Bax negative" phenotype.Lost of Bax expression is generally thought to be beneficial to the development of the tumor and chemo-resistance.However, some patients with "Bax-negative" MSI tumors have a better prognosis.The underlying molecular mechanism is not clear.We recently found that a unique Bax isoform (Bax△2) can be generated in combination of the deletion mutation and alternative splicing.
