目的 前脑NR2B亚基过表达的转基因模式动物,在脑片电生理上表现为LTP增强,在行为学上表现出较正常动物更强的空间学习和记忆能力.在啮齿类动物海马存在着与空间学习记忆能力密切相关的位置细胞(place cell),本研究重点考察NR2B转基因Long-Evans大鼠海马CA1区位置细胞的放电特性,并与正常动物相比较,从神经元活动水平探讨NR2B基因过表达对学习记忆能力影响的神经机制.方法 运用多通道在体记录技术,以回形跑道往返运动为行为模式,观察和记录海马CA1脑区群体神经元的放电活动和场电位活动状况,并重点分析位置细胞的反应特性.结果 (1)记录到的大多数位置细胞只有一个单方向的位置野,个别位置细胞存在编码双方向的位置野,且不同方向上的位置野在空间上并不重叠,在位置野内的放电频率也有所不同;(2)将回形跑道相对于周围整体环境旋转180°后,大部分位置细胞的位置野在跑道内会发生重排,但仍有一部分保持不变;(3)本底放电频率一致的位置细胞,当动物位于其位置野内时,NR2B过表达大鼠的位置细胞放电频率高于野生型大鼠(P<0.05).结论 NR2B过表达大鼠较正常大鼠在位置细胞频率编码上存在差异.
