目的讨论SPF级正常SD大鼠的骨髓有核细胞分类计数的参考值。方法收集2004年~2006年本中心所做实验对照用6周龄SD大鼠骨髓涂片共56例,雌雄各半,其中饲养28d20例,13周16例,26周20例。骨髓涂片制作方法为SD大鼠乙醚麻醉、腹主动脉放血处死后,进行骨髓涂片。采用Gimsa和Wrights混合染色30min左右。低倍电子显微镜下观察涂片,分类计数500个有核细胞。计数粒细胞系(包括粒母细胞,早幼粒细胞,中晚幼粒细胞,成熟粒细胞,嗜酸或嗜碱粒细胞),红细胞系(红母细胞,早幼红细胞,中幼红细胞,晚幼红细胞),淋系,计算各系细胞所占百分率及粒红比值。各指标均取平均值和标准差范围。结果饲养28dSD大鼠的粒系、红系、淋系细胞占有核细胞百分率和粒红比值在雄性分别为41.4±5.9,38.7±6.4,20.6±4.4和1.23±0.40;在雌性分别为42.5±7.1,36.5±6.6,20.8±4.2,1.11±0.29。各指标在饲养13周雄性分别为45.8±6.4,34.8±6.5,19.0±4.3,1.38±0.40;雌性分别为44.5±8.2,36.9±8.6,18.3±2.8,1.32±0.55。在饲养26周雄性分别为47.0±9.3,36.8±7.9,14.6±6.2,1.3±0.5;饲养26周雌性分别为43.6±10.7,38.9±7.4,16.2±4.3,1.2±0.5。结论大鼠粒红比值波动范围较大。随着大鼠鼠龄的增加,淋系细胞所占百分率略减少。
Objective To discuss the reference value of the classification and counting of bone marrow nucleated cells in SPF normal SD rats. Methods A total of 56 bone marrow smears from 6-week-old SD rats were collected from 2004 to 2006 in our center. There were 20 males and 28 females, 28 of them were bred for 28 days, 16 in 13 weeks and 20 in 26 weeks. Bone marrow smear production methods for the SD rat ether anesthesia, abdominal aorta bleeding after death, bone marrow smear. Mixed with Gimsa and Wrights for about 30 minutes. Observation under low electron microscope smears, classification counting 500 nucleated cells. Counting granulocytes (including granulocytes, promyelocytes, late promyelocytes, mature granulocytes, eosinophils or basophils), erythroid lines (red blood cells, premature erythrocytes, neutrophils, late Erythrocytes), lymph node, calculate the percentage of each line of cells and the ratio of grainy. The indicators are average and standard deviation range. Results The percentages of nucleated cells and the ratio of granulocytes to erythrocytes in gonorrhoeae, erythroid and lymphoid neoplasms of fed 28dSD rats were 41.4 ± 5.9, 38.7 ± 6.4, 20.6 ± 4.4 and 1.23 ± 0.40 respectively in males and 42.5 ± 7.1 , 36.5 ± 6.6, 20.8 ± 4.2, 1.11 ± 0.29. The indexes of the 13 weeks of feeding were 45.8 ± 6.4, 34.8 ± 6.5, 19.0 ± 4.3 and 1.38 ± 0.40 respectively; the females were 44.5 ± 8.2, 36.9 ± 8.6, 18.3 ± 2.8 and 1.32 ± 0.55, respectively. The 26-week-old males were 47.0 ± 9.3, 36.8 ± 7.9, 14.6 ± 6.2, and 1.3 ± 0.5, respectively. The 26-week-old females were 43.6 ± 10.7, 38.9 ± 7.4, 16.2 ± 4.3, and 1.2 ± 0.5, respectively. Conclusion The range of the ratio of grainy to grainy rats is larger. As rats grew, the percentage of lymphoid cells decreased slightly.
