HPLC-PDA and LC-tandem MS methods have been developed and validated for the simultaneous analyses of indolequinazoline-(IQAs) and quinolone alkaloids(QAs) in a herbal drug,"Evodiae Fructus".The bioactive markercompounds, evodiamine(1), rutaecarpine(2) and quinolone alkaloids were simultaneously determined in the 20 drug specimens that collected from Korea and China markets..HPLC analysis was performed on a reversed-phase phenyl-C6 column using solvent system of MeOH(A): 20mM NH4OAc(B): AcCN(C) using gradient flow detecting at UV 254nm.LC-ESI-MS/MS analysis was accomplished by multiple reaction monitoring(MRM) method after separation on a minibore phenyl-C6 column using 74% AcCN (5rnM NH4Ac).Contents of evodiamine(1) and rutaecarpine(2) were in the range between 0.12-1.14% and 0.07-1.02%, respectively, which together with other compounds, showed variable profiles depending on specimens.Clustering analysis was performed as an evaluating mean of the herbal drug.These methods would be useful as a crucial quality control of the herbal drug or the related products.
