目的:探讨骨髓间充质干细胞(BMSC)对脂多糖(LPS)诱导的小鼠内毒素血症的免疫调节作用。方法:选取体重30~40g的11~12周龄雄性昆明小鼠21只,随机分为3组:模型组、BMSC处理组、空白对照组,每组7只。模型组及BMSC处理组采用10mg/Kg LPS腹腔注射制备内毒素血症小鼠模型,空白对照组注射同等体积PBS;BMSC处理组在注射LPS1h后通过尾静脉注射体外扩增的BMSC(1×106/只),模型组和空白对照组注射同等体积PBS。24h后处死动物,留取各组组织标本。组织化学法检测肺脏和肝脏组织的炎性损伤程度并测定肺脏湿干比值;ELISA检测肺脏组织匀浆中炎性细胞因子IL-1β和TNF-α水平。
Objective: To investigate the immunomodulatory effect of bone marrow mesenchymal stem cells (BMSCs) on lipopolysaccharide (LPS) -induced mouse endotoxemia. Methods: Twenty-one male Kunming mice (11-40 weeks old) weighing 30-40 g were randomly divided into three groups: model group, BMSC group and blank control group, with 7 mice in each group. The model group and the BMSC treatment group were injected with 10mg / Kg LPS intraperitoneal injection of endotoxemia mice model, the blank control group injected with the same volume of PBS; BMSC treatment group after injection of LPS1h after injection through the tail vein in vitro amplification of BMSC (1 × 106 / Only), model group and blank control group injected the same volume of PBS. The animals were sacrificed 24h later, and the tissue samples of each group were collected. Histochemical method was used to detect the degree of inflammatory injury in lung and liver tissues and lung wet / dry ratio. The level of inflammatory cytokines IL-1β and TNF-α in lung homogenates were detected by ELISA.
