A special microfluidic chip for organophosphorus pesticides detection is made based on enzyme inhibition method.The chip is fabricated from polydimethylsiloxane(PDMS).An inner rib type micro mixing chamber is arranged in microfluidic chip for sample reagent and chromogenic agent mixing.The surface of the mixing chamber fixed with acetylcholinesterase is used for inhibiting enzyme reaction in the mixing process.A photoelectric detection area is made behind the mixing chamber.In order to increase the detection optical path,a reflection probe and a total reflection lens are arranged above and below the detection area respectively.The are connected with optical source and spectrometer.Absorbance and spectral changes were recorded using the software provided by the manufacturer of the spectrometer,which was controlled by a portable computer.Experiment results show that when the concentration of organophosphorus pesticide is at range of 3.05×10-6~4.05×10-4mol/L,the absorbance measured by reflection probe have a good linearity of 0.9912.
