Getting rid of ensemble average,experiments on single molecule level are informative and powerful to study the dynamics of biomolecutar system.In order to leam dynamic properties of biomolecules in a wide time window,we built up a scanning confocal microscopy system.With piezo stage controlled by LabVIEW software,we were able to scan sample areas and then locate single fluorescent molecules immobilized on the glass slide surface.This technique is prospective to extract dynamic information on timescales ranging from tens of microseconds to tens of seconds.We tested our apparatus using single molecule bleaching experiment to verify its capability in spatial resolution for single molecule detection and manipulation.
