  Mesenchymal stem cells (MSCs) are the adult progenitor cells for the mesodermal lineages of adipocytes, osteoblasts, chondrocytes, and fibroblasts.Recent reports show that these stem cells possess strong immunomodulatory properties-including expansion of immunomodulatory leukocytes such as regulatory T lymphocytes and tolerogenic dendritic cells-that are clinically relevant.Interestingly,mechanistic studies have revealed that many similarities exist between MSC immunomodulation and cancer immune evasion.Our lab has a long-standing interest in delineating mechanisms of action for these properties.We demonstrated that human MSC-secreted hepatocyte growth factor (HGF) can expand CD14-CD11b+CD33+ myeloid-derived suppressor cells (MDSCs) as well as rapidly convert CD14+ monocytes into an immunomodulatory, interleukin (IL)-10-secreting phenotype.Most recently, we have found that MSCs can suppress IL-17-expressing CD4 cells (Th17) responses through a secreted factor and STAT3 to modulate surface expression of Programmed Death-Ligand 1 (PD-L1) , a potent ligand of the immune checkpoint Programmed Death-1 (PD-1).Our data suggest that mechanistic understanding of human MSC immunomodulation may yield targets for therapeutic application in both immune-mediated diseases as well as cancer.
