Dopamine (DA) has been reported to involve in the regulation of insulin secretion of pancreatic islets and protection of gastrointestinal (GI) mucosa.However, where the DA comes from is controversial.The present study investigated the source of pancreatic DA in rat by means of high-performance liquid chromatographymass spectrometry (HPLC-MS), immunofluorescence and western blot.The result from HPLC-MS indicated that pancreatic tissue contained a certain amount of DA (108.3±15.8 ng/mg tissue).Tyrosine hydroxylase positive immunoreactivity was widely distributed in the cells of pancreatic acinus and some nerve fibers surrounding the islets and acinus.The expression of TH protein was much higher in exocrine pancreas than in isolated islet.In conclusion, DA might be synthesized in acinar cells and secreted to the GI tract and islets through solinocrine and/or paracrine.
