Capsid-associated tegument components have been identified in alpha-and beta-herpesviruses to play essential roles in viral DNA-packaging.Whether and how such tegument components exist in gamma-herpesviruses have been a mystery. Here we report a 4(A)resolution cryoEM reconstruction of the Kaposis sarcoma-associated herpesvirus (KSHV) virion. The reconstruction reveals for the first time the structures of the capsid and capsid-associated tegument proteins at atomic details.
