本文以猕猴桃病毒病为研究对象,对侵染猕猴桃的病毒种类进行了鉴定,已发现的病毒有猕猴桃病毒A(Actinidia virus A,AcVA)、猕猴桃病毒B(Actinidia virus A,AcVB)和苹果茎沟病毒(apple stem grooving virus,ASGV)。同时采用siRNA测序技术鉴定出一种侵染猕猴桃的欧洲山棒环斑病毒属(Emaravirus)新病毒,根据拼接序列设计引物扩增获得其全基因组序列,其基因组由4条负义单链RNA组成,各RNA分别含1个开放式阅读框,其中RNA1大小为7061个核苷酸(nt),推测编码2303个氨基酸的复制相关蛋白;RNA2由2267个核苷酸组成,编码含653个氨基酸的糖蛋白;RNA3为1678个核苷酸,编码310个氨基酸的核衣壳蛋白;RNA4大小为1664个核苷酸,编码379个氨基酸的功能未知蛋白。该病毒的RNA1~4与redbud yellow ringspot virus(RYRV)相应RNA相似性最高,分别为57.5%(RNA3)—65.7%(RNAl),所推测的编码蛋白相似性分别为65.1%、48.3%、55.7%和58.9%;且在各RNA的3’和5’端均具有由15个nt和17个nt组成的高度保守的反向重复序列。根据其序列特点,初步确定该病毒为Emaravirus属的一个新的病毒种。
