灯盏花[Erigeron breviscapus (Vant.) Hand.-Mazz]为菊科飞蓬属多年生植物,是云南省特有的药用植物之一.以自交不亲和的灯盏花为材料,灯盏花转录组的unigene序列设计引物,通过RACE的技术,从灯盏花的花序中分离得到了一个SRK基因的cDNA全长,该基因全长为2891bp,包括2634bp的ORF,116bp的5'UTR和138bp的3'UTR,编码878个氨基酸,其氨基酸序列与菊科千里光的SRK基因有较高的同源性.生物信息学分析表明SRK蛋白具有跨膜域,激酶结构域,ATP结合位点等.这些都是SRK蛋白的典型特征.RT-PCR结果表明:灯盏花SRK基因的表达不仅仅存在柱头中,叶片及茎等营养器官中也有表达.灯盏花SRK基因的克隆和表达模式分析为研究菊科植物孢子体自交不亲和奠定了一定的基础.
