TET2 is a noveltumor suppressor gene involved in several hematological malignancies of myeloid and lymphoid origin.Besides loss-of-function mutations and deletions,hypermethylation of the CpG island at the TET2 promoter was found in human cancer.The aim of the present study was to determine if promoter hypermethylation may be a mechanism of TET2 inactivation in a group of acute myeloid leukemia(AML)cases.In the present study,quantitative reverse transcriptase-PCR(qRT-PCR)and methylation-specific polymerase chain reaction(ms-PCR)were used to analyze TET2 mRNA expression and methylation in 38 AML patients,including M1(3/38),M2(5),M3(6),M4(5),M5(17),and M6(2),respectively.In one of the 38 AML patients,methylation of TET2 promoter region was detected by msPCR.
