Research on the effect of Neuromedin U2 receptor activated by the citrus flavonoids

  Objective To identify the agonists for Neuromedin U 2 receptor by using NMU2R cell line which was established by cotransfecting HEK293 with the NMU2R plasmid (NMU2R/pCDNA3.1 +) and reportergene plasmid (3×MRE/3×CRE/SRE-Luciferase/pGL3).Methods Plate NMU2R cells in growth medium with 3% FBS in a 96-well format and incubate the cells at 37 ℃ in a CO2 incubator for 12 hours.Add the citrus flavonoids and Forskolin solutions to each well containing cells and medium.Incubating the cells at 37 ℃ in a CO2 incubator for 6-8 hours.Add an equal volume of 0.5×Bright-GloTM solution, then test the value of a luciferase signal.Establish the activation effect of the various citrus flavonoids to Neuromedin U 2 receptor by the value of a luciferase signal.Results (1) Naringin, Naringenin and Tangeretin can not activate Neuromedin U 2 receptor.(2) Hesperidin and Nobiletin can activate Neuromedin U 2 receptor.(3) The EC50 values of Hesperidin and Nobiletin for the receptor were 319 μM and 6 μM.Conclusion Hesperidin and Nobiletin can be the agonists of Neuromedin U 2 receptor.
