目的痘苗病毒作为疫苗载体具有接种途径简单、高滴度、低成本等优点,重组痘苗病毒投入使用之前进行理化特性试验,对重组痘苗病毒的生产、运输和保存有一定重要意义。方法将重组痘苗病毒rVTT-JEVE作热、酸、碱、乙醚、氯仿、胰蛋白酶、超声波以及紫外线处理,然后接种到BHK-21细胞上,观察rVTT-JEVE毒价的变化,分析理化因素对rVTT-JEVE的影响。结果在pH3~9范围内,酸碱度变化不会引起rVTT-JEVE毒价的显著变化。rVTT-JEVE经50℃处理60min、30min和60℃处理15min可被灭活;对5%的氯仿敏感;经终浓度0.5%的胰蛋白酶37℃作用1h,病毒毒价降低2个滴度;对25%的乙醚不敏感;经40KHz超声波处理60min和80KHz处理30min即可灭活;30W紫外灯垂直距离10cm照射15min、30cm照射30min即可灭活。结论 rVTT-JEVE耐热、酸、碱、乙醚、氯仿及胰蛋白酶,对超声波、紫外线较敏感。
Objective The vaccinia virus has the advantages of simple vaccination route, high titers, low cost and the like. The vaccinia virus has the physical and chemical properties test before it is put into use, and has certain significance for the production, transportation and preservation of the recombinant vaccinia virus. Methods The recombinant vaccinia virus rVTT-JEVE was treated with heat, acid, alkali, ether, chloroform, trypsin, ultrasonication and UV light and then inoculated onto BHK-21 cells to observe the changes of the virulence of rVTT-JEVE. -JEVE effect. Results In the range of pH 3-9, changes in pH did not cause significant changes in the virulence of rVTT-JEVE. rVTT-JEVE could be inactivated after treatment with 50 ℃ for 60min, 30min and 60 ℃ for 15min, sensitive to 5% chloroform, and the virulence of virus decreased by 2 titers after treated with 0.5% trypsin at 37 ℃ for 1h. 25% of the ether is not sensitive; 40KHz ultrasonic treatment 60min and 80KHz treatment 30min inactivation; 30W UV lamp vertical distance 10cm irradiation 15min, 30cm irradiation 30min inactivation. Conclusions rVTT-JEVE is resistant to heat, acid, alkali, ether, chloroform and trypsin and is sensitive to ultrasound and ultraviolet light.
