Analysis of the genomic sequence of the self-flocculating yeast SPSC01 and comparative transcriptomi

  Saccharomyces cerevisiae has been widely used for bioethanol production.We have developed a flocculating yeast strain SPSC01 which has stable flocculation property and excellent tolerance against various environmental stresses.In this study,the genomic sequence of SPSC01 was analyzed.The estimated genome size of SPSC01 is 15.35 Mb,with 5315 coding genes and the gene average length is about 1531 bp.Several Single Nucleotide Polymorphisms(SNPs)were found in SPSC01 compared to the model laboratory yeast S288c,including the SNP in certain transcription regulator.Under the condition of continuous ethanol fermentation process,differential gene expression profiling of the SPSC vs the SPSC FLO1Δ mutant was analyzed.Among 802 differentially expressed genes,114 genes were upregulated,and 62 genes were downregulated.Some genes with elevated transcription levels were confirmed to be involved in the improved stress tolerance of SPSC01.These results provide basis for the second generation of ethanol production.
