In South Asian countries,oils from mustard (Brassica rapa) are important sources of vegetable fat,but zero erucic acid mustard is not available in those countries.In this study,to develop erucic acid free mustard,the high erucic acid B.rapa cultivars,Tori-7 (Toria) and BARI sarisha 14 (Yellow sarson),were backcrossed as recurrent parents to a canola quality B.napus Kirariboshi.Since it has been reported that the erucic acid free trait of B.napus comes from a mutation in two copies of the fatty acid elongase 1 (FAE1) gene,we designed dCAPS primer pairs,which can amplify A-genome FAE1 gene separately from C-genome FAE1 gene,and furthermore distinguish between the mutated and wild type FAE1 genes.
