  115 bacterial strains were screened from soil,rotten leather and water of a tannery,Foshan,China.According to the size of hydrolyzed circle,ability of degrading Nile tilapia skin and collagenase activity,Sub-1 had high collagenase activity.Based on the bacterial characteristics,culture conditions,physiological and biochemical experiments,and 16S rRNA sequence,Sub-1 was identified as Bacillus subtilis.Sub-1 had high collagenase activity in degrading fresh fish skin into was less than 5 kDa soluble peptides.Gelatin zymography analysis indicated it could secret alkaline metalloproteinases whose maximal gelatinolytic activities were attained at pH 7.5 and 45℃,Ca2+,Mn2+,Mg2+ and Zn2+ ions enhanced the collagenase activity,whereas Cu2+,Hg2+,Al3+,Co2+,Fe2+ ions suppressed it.Single factor experiment suggested that optimal fermentation period was 40h,optimal carbon source was sucrose and optimal nitrogen source was tryptone.Orthogonal test showed optimized substrate components were 1.5%sucrose,1.5%tryptone,0.2%gelatin and 0.25%yeast.Taken together,strain Sub-1 could be potential for collagenase production and fishery waste degradation.
