Modulation of delayed rectifier potassium current by N/OFQ via Ca2+-CaM-CaMKII

  Nociceptin/orphanin FQ (N/OFQ) plays an important role in a variety of physiological functions including learning and memory by exerting effects on different ion channels.Here we showed for the first time that the effects of Ca2+-CaM-CaMKⅡ on the inhibition of IK by N/OFQ in acutely dissociated rat cerebral parietal cortical neurons.Our previous reported that N/OFQ inhibited delayed rectifier potassium current (IK) which were associated with PKC and ERK1/2 pathway.In this present study, we used the conventional whole cell patch-clamp technique.We wondered if Ca2+-CaM-CaMKⅡ is involved in the inhibitive effect of N/OFQ on IK.We found that W-7, a specific inhibitor for CaM, and KN-62, an inhibitor of CaMKII, attenuated the inhibitory effect of N/OFQ on the IK.These data suggest that the Ca2+-CaM-CaMKII pathway, at least in part, mediates the inhibitory effect of N/OFQ on the IK in acutely dissociated rat cerebral parietal cortical neurons.
