Se deficiency affected the mRNA expression of inflammatory factors and twenty-five selenoprotein gen

  The aim of the present study was to investigate effects of Se deficiency (SeD) on the mRNA expression of inflammatory factors and 25 selenoprotein genes in kidney of broiler chicks.150 1-day-old broiler chicks were randomly assigned to two groups fed with a low-Se diet (SeD group,0.033 mg/kg Se)or adequate Se diet(C group,0.2 mg/kg Se).The levels of uric acid (UA)and creatinine (Cr)in the serum,the mRNA levels of 6 inflammatory factors and 25 selenoprotein genes in kidney were determined as the symptom of SeD occurred.The results showed that the contents of UA and Cr in the serum were significantly increased in the SeD group.The mRNA levels of inflammatory factors (NF-κB, iNOS, COX-2, TNF-α)were significantly increased in SeD group.Meanwhile, the mRNA levels of PTGEs and HO-1 were not significantly changed in SeD group.In addition,25 selenoprotein transcripts displayed a ubiquitous expression in kidney of chicks.The mRNA levels of 14 selenoprotein genes (Dio1, Dio2, GPx3, Sepp1, SelH, SelI, SelK, Sepn1, SelO, SelW, Sel15, SelT, SelU and SelS)were decreased and 9 selenoprotein genes (GPx1, GPx2, GPx4, SelPb, TrxR1, TrxR2, TrxR3, SPS2 and SelM)were up-regulated in the SeD group,but the changes of Dio3 and Selx1 mRNA levels were not significant (P> 0.05).The results indicated that Se deficiency induced the abnormal renal functions.The change of the inflammatory factors mRNA played an important role in the abnormal renal functions induced by SeD.25 selenoprotein genes showed a different expression in SeD group.
