At first,we optimized separation protocol of membrane proteins of Aspergillus fumigates by 2D Fluorescence difference gelelectrophoresis (2D DIGE).We found that membrane proteins were dissolved well in followed lysis buffer:7Murea,4%CHAPS,2Mthiourea,1%ASB14,30mMTris,2% TritonX-100,1%DTT,1%IPG buffer.According to this effective 2D DIGE procedure,weresearched the differential expression of membrane proteins among the wild,pmt1 and pmt2 whose mannose transferase wasknocked out.
