Flavonoids such as proanthocyanidins have been known to reduce the adverse consequences of excessive alcohol and iron on the liver,with procyanidin being an effective example.Procyanidins have currently gathered increasing attention and are becoming the prime center of study owing to their antioxidant properties.Procyanidins are naturally occurring compounds that have medicinal effects and are consumed in large amounts in foods such as legumes,fruits,grains,and nuts.Purpose:The liver can be a cause of cardiac disease.In this regard,a better understanding of cardiohepatic interactions plays a vital role in the management of hepatic and cardiac dysfunctions.So,managing liver disease could help prevent certain cardiac anomalies.This study sought to investigate the anti-inflammatory properties of these powerful antioxidants called procyanidins in stopping the development,or further progression of Alcoholic Liver Disease(ALD),by analyzing the compounds’ natural activities on hepatic injury triggered by excessive ethanol and iron consumption.Methods:Fifty-one rats(Wistar,male)were randomly divided into four groups: A,B,C,and D.The first being the control Group A(normal diet);the second was model Group B(high alcohol 8-12 m L/kg/d along with iron 1000 mg/kg diet);the third group being low dose procyanidin Group C(Group B diet together with 60 mg/kg/d of procyanidin)and the final group being high dose procyanidin Group D(Group B diet together with 120 mg/kg/d of procyanidin).The rats were sacrificed after 12 weeks to detect liver damage due to pathological changes.Biochemical markers serum aspartate aminotransferase(AST),alanine aminotransferase(ALT),and gamma-glutamyl transpeptidase(GGT)along with serum and liver tissue triglyceride(TG),high-density lipoprotein cholesterol(HDL-C),total cholesterol(TC),and low-density lipoprotein cholesterol(LDL-C)were measured spectrophotometrically by the automatic biochemical analyzer.Reverse Transcription Polymerase Chain Reaction(RT-PCR)was used to assess the NFκB and IκB m RNA expression levels.Western blotting was used to evaluate the NFκB p65 and IκB protein expression levels,while ELISA was used to identify serum inflammatory factors tumor necrosis factor-alpha(TNF-α),Interleukin(IL)-6,IL-10,and IL-4.Results:The result revealed that procyanidin displayed significant hepatoprotective effects.In Group B that was treated with ethanol and iron,levels of total cholesterol(TC),serum triglyceride(TG),liver triglyceride,and low-density lipoprotein cholesterol(LDL-C)were increased;and significantly decreased by 21%,16%,20%,45%,respectively(P < 0.05)in the120mg/kg procyanidin Group D.Meanwhile,procyanidin significantly reversed the increased level of serum inflammatory factors like tumor necrosis factor-alpha(TNF-α),Interleukin-6,Interleukin-10,and Interleukin-4.Nuclear factor κB(NFκB)triggering was suppressed by procyanidin.Conclusion:The study results indicated that excessive quantities of alcohol and iron cause liver damage due to increased release of cytokines,inflammatory markers,and low pathological scores.Large amounts of procyanidin are shown to remedy the effects of liver damage by reducing the release of cytokines,TG,TC,and LDL-C while improving the pathological score of the liver through maintenance of its integrity.The outcome underlined the physiological damages of consuming alcohol and diets with high iron contents but proved that procyanidins could be effective in reversing these impacts and preserving liver health.
