目的探讨在不同诱导因素条件下,不同来源的阿萨希毛孢子菌(Tricho sporon asahii,T.asahii)菌丝诱导结果的差异。方法 11株不同来源的T.asahii,选用RPMI-1640液体培养基、YPD液体培养基、吐温酵母肉汤(TYB)培养基和50%(v/v)胎牛血清,分别在15℃、25℃和37℃条件下培养,于1h、2h、3h、6h、12h、24h计数菌丝生成率。结果 T.asahii 15℃菌丝生成率均值为(0.15±0.42)%,25℃为(5.75±3.48)%,37℃为(33.81±15.39)%,差异有统计学意义(P值均<0.05)。RPMI-1640培养基诱导菌丝生成率均值为(46.24±25.50)%,YPD液体培养基为(36.28±21.85)%,TYB培养基为(33.93±21.29)%,50%(v/v)胎牛血清为(18.60±14.58)%,组间比较有显著统计学差异(F=6.29,P=0.0013),组内比较有显著统计学差异(F=21.80,P=0.0000)。随着诱导时间的延长,各菌株菌丝生成率也随之上升。结论综合比较各诱导因素,RPMI-1640培养基、37℃诱导24h可使大多数T.asahii菌株由孢子相获得较纯的菌丝相,而菌株来源的不同也是影响诱导结果差异的重要因素之一。
Objective To investigate the differences in the induction of mycelia of Tricho sporon asahii (T.asahii) from different sources under different inducing factors. Methods Eleven different strains of T.asahii were selected and cultured in RPMI-1640 liquid medium, YPD liquid medium, TYB medium and 50% (v / v) fetal bovine serum at 15 ℃, 25 ℃ and 37 ℃ under the conditions of culture, in 1h, 2h, 3h, 6h, 12h, 24h mycelial growth rate was counted. Results The average mycelial growth rates of T.asahii at 15 ℃ were (0.15 ± 0.42)%, (5.75 ± 3.48)% at 25 ℃ and (33.81 ± 15.39)% at 37 ℃, respectively ). The mean mycelial induction rate of RPMI-1640 medium was (46.24 ± 25.50)%, that of YPD liquid medium was (36.28 ± 21.85)%, that of TYB medium was 33.93 ± 21.29%, and that of 50% (v / v) Bovine serum was (18.60 ± 14.58)%, there was a significant statistical difference between the two groups (F = 6.29, P = 0.0013). There was significant difference between the two groups (F = 21.80, P = 0.0000). With the extension of induction time, the mycelial growth rate of each strain also increased. Conclusion The results showed that most T.asahii strains obtained relatively pure mycelial phase from spore phase in RPMI-1640 medium at 37 ℃ for 24 h. However, the origin of different strains was also an important factor affecting the difference of induced results one.
