目的:探讨加味导赤散与维生素B12联合治疗复发性口腔溃疡(ROU)动物模型的作用及其对血清IL-6及TNF-α的影响。方法:采用免疫方法建立ROU动物模型。48只SD大鼠随机分为正常组、模型组、左旋咪唑组(对照组)、低剂量加味导赤散加维生素B12组(治疗组1)、中剂量加味导赤散加维生素B12组(治疗组2)、高剂量加味导赤散加维生素B12组(治疗组3),每组8只。对照组用左旋咪唑片灌胃,治疗组分别用低、中、高剂量的加味导赤散与维生素B12一起灌胃,观察记录溃疡变化情况,用酶联免疫法(ELISA)测定大鼠造模前后及灌胃后血清IL-6及TNF-α含量。结果:1.实验大鼠于第3次注射抗原乳化剂1周后开始出现口腔溃疡。2.对照组与治疗组2、组3的大鼠口腔溃疡分别较模型组发生数目明显减少,持续时间明显缩短,间隔时间明显延长(P<0.05)。3.模型组IL-6及TNF-α水平较正常组明显升高(P<0.05),而对照组与2、3治疗组较溃疡组下降(P<0.05)。结论:加味导赤散加维生素B12可能通过下调IL-6及TNF-α水平促进口腔溃疡愈合,减少溃疡复发。
Objective: To investigate the effect of Jiawei Daochi Powder and Vitamin B12 in the treatment of recurrent oral ulcer (ROU) animal model and its effect on serum IL-6 and TNF-α. Methods: The animal model of ROU was established by immunization. Forty-eight SD rats were randomly divided into normal group, model group, levamisole group (control group), low-dose Jiawei Daoguci Granule B12 treatment group (treatment group1) and middle-dose Jiawei Dickensacci Vitamin B12 group Group 2), high-dose flavored Vasopressin plus vitamin B12 group (treatment group 3), 8 in each group. The control group was treated with levamisole gavage. The treatment group was administered with low, medium and high doses of Jiawei Daochi Powder together with vitamin B12, the change of ulcer was observed and recorded, and the rat model was established by enzyme-linked immunosorbent assay (ELISA) Serum levels of IL-6 and TNF-α were measured before and after gavage. The experimental rats began oral ulcers after the third injection of antigen emulsifier for 1 week. The number of oral ulcer in the control group and the treatment group 2 and group 3 was significantly reduced compared with the model group, the duration was significantly shortened, and the interval time was significantly longer (P <0.05). The levels of IL-6 and TNF-α in the model group were significantly higher than those in the normal group (P <0.05), while those in the control group and the 2,3 treatment group were lower than those in the ulcer group (P <0.05). Conclusion: Jiawei Daoshi Granula B12 may promote the healing of oral ulcer and reduce the recurrence of ulcer by decreasing the levels of IL-6 and TNF-α.
