目的分析钉螺软体组织中的蛋白水解酶。方法以明胶作底物,利用明胶-SDS-聚丙烯酰胺凝胶电泳(PAGE)分离钉螺软体组织蛋白质,于不同缓冲液和酶抑制剂中进行孵育,分析和鉴定其中的蛋白水解酶。结果钉螺软体组织中存在降解明胶的蛋白水解酶,其最适pH值为碱性。金属蛋白酶抑制剂乙二胺四乙酸二钠(EDTA)可抑制其活性。电泳分离获得有活性的酶蛋白。结论钉螺软体组织中存在被EDTA抑制的蛋白水解酶。
Objective To analyze the proteolytic enzymes in Oncomelania hupensis. METHODS: Gelatin was used as a substrate. Proteins of Oncomelania snail were separated by gelatin SDS-polyacrylamide gel electrophoresis (PAGE) and incubated in different buffers and enzyme inhibitors to analyze and identify the proteolytic enzymes. Results There was gelatin degrading proteolytic enzyme in the soft tissue of Oncomelania hupensis, and its optimum pH value was alkaline. Metalloproteinase inhibitor disodium edetate (EDTA) inhibits its activity. Electrophoresis separation of active enzyme protein. CONCLUSIONS There is a protease that is inhibited by EDTA in the snail soft tissue.
