目的:研究中药复方活血通络汤对过氧化氢所致人脐静脉内皮细胞(HUVEC)损伤保护作用的机制。方法:制备兔含药(活血通络汤)血清,HUVEC复苏和培养后,建立氧化损伤模型,分组后进行处理,倒置显微镜下观察其形态学变化,测定培养液上清LDH、MDA含量及对细胞凋亡的影响。结果:活血通络汤含药血清低、中、高剂量组与模型组比较能降低HUVEC细胞上清LDH及MDA的含量,减少细胞凋亡现象。结论:活血通络汤对过氧化氢所致人脐静脉内皮细胞(HUVEC)损伤有明显的保护作用。
Objective: To study the protective mechanism of Huoxue Tongluo Decoction, a compound traditional Chinese herbal medicine, on the damage of human umbilical vein endothelial cells (HUVEC) induced by hydrogen peroxide. METHODS: Rabbit serum containing drug (Huoxuetongluo Decoction) serum was prepared. HUVECs were resuscitated and cultured. Then, oxidative damage models were established. They were grouped and processed. Morphological changes were observed under an inverted microscope to determine the content of LDH and MDA in the culture supernatant. The effect of apoptosis. RESULTS: Compared with the model group, the serum containing Huoxuetongluo decoction lowers the content of LDH and MDA in the supernatant of HUVEC cells and reduces the apoptosis. Conclusion: Huoxue Tongluo Decoction has obvious protective effect on hydrogen peroxide-induced human umbilical vein endothelial cells (HUVEC) injury.
