目的:研究八角莲对大鼠肝组织超氧化物歧化酶(superoxide.dimutase,SOD)、丙二醛(MDA)、大鼠活性氧簇(ROS)、丙氨酸转氨酶(ALT)、天门冬氨酸氨基转移酶(AST)、γ-谷氨酰转移酶(γ-GT)及肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的影响,探讨八角莲致大鼠肝脏损害的毒性机制。方法:分别给予SD大鼠八角莲水提液与醇提液连续灌胃3 d后,取正常组及八角莲给药组(3.50和2.96 g·kg~(-1))大鼠肝组织,制成10%肝匀浆,按试剂盒方法测定肝组织中SOD、MDA、ROS活性和ALT、AST、γ-GT含量,计算SOD/MDA;采用双抗体夹心ABC-ELISA法测定大鼠肝组织TNF-α、IL-6含量。结果:与正常组比较,八角莲水提组与醇提组大鼠肝组织SOD下降,MDA上升,SOD/MDA下降,ROS值上升(P<0.05);ALT、AST、γ-GT、TNF-α、IL-6含量升高(P<0.05)。结论:大鼠灌胃给予八角莲后,可对肝脏产生明显的毒性,其机制可能与影响自由基及炎症因子有关。
OBJECTIVE: To study the effects of octagood on the expression of superoxide dismutase (SOD), malondialdehyde (MDA), reactive oxygen species (ROS), alanine aminotransferase (ALT), aspartate (AST), γ-glutamyl transferase (γ-GT), tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) Toxicity of rat liver damage. Methods: SD rats were treated with aqueous extract of octagonal lotus and alcohol extract continuously for 3 days. The rats in the normal group and the ocuporterine group (3.50 and 2.96 g · kg -1) The activity of SOD, MDA and ROS, the content of ALT, AST and γ-GT in the liver tissue were determined by kit method, and the SOD / MDA was calculated. The liver tissue of rat was measured by double antibody sandwich ABC-ELISA TNF-α, IL-6 content. Results: Compared with the normal group, the activity of SOD, the content of MDA, the content of SOD and the content of ROS in the liver of SD group and alcohol group decreased (P <0.05) α, IL-6 levels increased (P <0.05). Conclusion: Oral administration of octopsis ganoderma after intragastric administration can produce obvious toxicity to the liver, which may be related to the effects of free radicals and inflammatory factors.
