目的研究96例胃癌组织和胃癌细胞株(MKN-28、SGC-7901、BGC-823、HGC-27)中胃肠富集Krüppel样因子(Gut-enriched Krüppel-like factor,GKLF)、肠道富集Krüppel样因子(Intestinal-enriched Krüppel-like factor,IKLF)的表达及意义。方法荧光实时定量PCR法检测胃癌组织和胃癌细胞株中GKLFmRNA及IKLF mRNA的水平;分别采用免疫组织化学法和Western blot检测胃癌组织和细胞株中GKLF、IKLF蛋白的表达。结果胃癌组织中GKLF mRNA水平低于正常胃黏膜,IKLF mRNA水平高于正常胃黏膜(P均<0.0001),GKLF mRNA、IKLF mRNA的水平与胃癌分化程度无关。胃癌组织中GKLF蛋白阳性率显著低于正常胃黏膜,IKLF蛋白阳性率则显著高于正常胃黏膜(P均=0.001);GKLF、IKLF蛋白表达与胃癌患者的年龄、性别、分化程度、Lauren分型无关,但与淋巴结是否有转移及pTNM分期相关(P=0.001)。胃癌细胞株中GKLF mRNA及蛋白水平低于人正常胃上皮细胞株GES-1,IKLF mRNA水平及蛋白则高于细胞株GES-1(P均<0.05)。结论 GKLF、IKLF可能参与了胃癌的发生、发展,检测GKLF、IKLF的表达有助于判断胃癌患者病情程度和预后。
Objective To investigate the effects of Gut-enriched Krüppel-like factor (GKLF) in 96 gastric cancer tissues and gastric cancer cell lines (MKN-28, SGC-7901, BGC-823 and HGC-27) Expression of Intestinal-enriched Krüppel-like Factor (IKLF) and Its Significance. Methods The levels of GKLF mRNA and IKLF mRNA in gastric cancer and gastric cancer cell lines were detected by real-time fluorescence quantitative PCR. The expressions of GKLF and IKLF protein in gastric cancer tissues and cell lines were detected by immunohistochemistry and Western blot respectively. Results The level of GKLF mRNA in gastric cancer tissue was lower than that in normal gastric mucosa, and the level of IKLF mRNA was higher than that in normal gastric mucosa (P <0.0001). The levels of GKLF mRNA and IKLF mRNA were not correlated with the differentiation of gastric cancer. The positive rate of GKLF protein in gastric cancer was significantly lower than that in normal gastric mucosa, while the positive rate of IKLF protein was significantly higher than that in normal gastric mucosa (P = 0.001). The expression of GKLF and IKLF was significantly correlated with the age, sex, differentiation, Lauren score But not with lymph node metastasis and pTNM stage (P = 0.001). The level of GKLF mRNA and protein in gastric cancer cell lines was lower than that in human normal gastric epithelial cell line GES-1. The level of IKLF mRNA and protein in gastric cancer cell line was higher than that in cell line GES-1 (all P <0.05). Conclusions GKLF and IKLF may be involved in the occurrence and development of gastric cancer, and the expression of GKLF and IKLF may be helpful to judge the severity and prognosis of gastric cancer.
