目的建立一种三维血管瘤血管生成体外培养模型。方法将新鲜的增殖期血管瘤手术标本置于纤维蛋白凝胶,以双层夹心法建立三维血管瘤血管生成体外培养模型。结果血管瘤组织培养2~3d后芽生出细小血管,后呈枝丫状生长,至第8~9天长成树枝状血管,分叉交叠,之后血管生长渐缓停滞。组织周边新生树枝状结构经石蜡切片HE染色、血管内皮细胞特异性标志物第Ⅷ因子相关抗原检测及电镜观察,鉴定为血管。结论该模型的成功建立有助于血管瘤血管生成机制及抗血管生成药物的研究。
Objective To establish a three-dimensional angiogenesis model of angiogenesis in vitro. Methods Fresh proliferating hemangiomas were placed on fibrin gels. A three-dimensional hemangioma angiogenesis in vitro culture model was established by double-layer sandwich method. Results Hemangiomas grew finely 2-3 days after budding, and then grew into branches. After 8 ~ 9 days, they grew into dendritic vessels and then branched into two branches. After that, the blood vessels grew slowly. The newborn dendritic structure around the tissue was stained by paraffin section, vascular endothelial cell specific marker Ⅷ factor-related antigen test and electron microscope observation, identified as blood vessels. Conclusion The successful establishment of this model contributes to angiogenesis and angiogenesis.
