目的:建立及优化人宫颈癌组织蛋白质提取方法,为下一步寻找差异表达的蛋白质并早期诊断宫颈癌提供新的思路。方法:利用一步法和三步法提取宫颈癌组织蛋白质,并分别用Bradford法测定相应样品总蛋白质浓度,并进行双向丙烯酰胺凝胶电泳分离,观察两种方法提取样品蛋白质含量及2-DE蛋白质图谱高分辨率的情况。结果:三步法提取的宫颈癌组织蛋白质含量较一步法有明显的增高,2-DE蛋白质图谱较一步法清晰、分辨率高。结论:与一步法相比较,三步法具有高蛋白质浓度、高分辨率、高清晰度的二维电泳图谱的优点,利于全面准确地进行蛋白质学研究。
OBJECTIVE: To establish and optimize the method of protein extraction from human cervical cancer tissue, and to provide a new idea for further searching for differentially expressed proteins and early diagnosis of cervical cancer. Methods: The protein of cervical cancer tissues was extracted by one-step and three-step method. The total protein concentration of the corresponding samples was determined by Bradford method and separated by two-dimensional acrylamide gel electrophoresis. The protein content and 2-DE protein Atlas of high resolution. Results: Compared with the one-step method, the protein content of cervical cancer tissue extracted by three-step method was significantly higher than that of one-step method. The protein profile of 2-DE was more clear and the resolution was higher. Conclusion: Compared with one-step method, the three-step method has the advantages of high protein concentration, high resolution and high-resolution two-dimensional electrophoresis, which is conducive to a comprehensive and accurate protein study.
