目的:探讨外胚间充质干细胞(EMSC)向树突状细胞(DCs)分化的能力。方法:利用基因重组大鼠GM-CSF和IL-3连续诱导17 d,在第19天加入基因重组大鼠TNF-α继续诱导,观察其形态、超微结构、表面标记、混合淋巴细胞反应、IL-12分泌功能等,以SD大鼠骨髓间充质干细胞向DC分化作为对照。结果:经过诱导的EMSC出现细长伪足和毛刺,变为半贴壁状态;扫描电镜显示长的细胞突起和许多毛刺;其表面标记与骨髓来源的DC相似;混合淋巴细胞反应证实其可以刺激淋巴细胞增殖;诱导后的EMSC有分泌IL-12功能。结论:EMSC在体外经诱导可向DC分化。
Objective: To investigate the ability of ectomesenchymal stem cells (EMSCs) to differentiate into dendritic cells (DCs). Methods: GM-CSF and IL-3 were induced continuously for 17 days in genetically modified rats, TNF-α was induced on the 19th day and the morphology, ultrastructure, surface markers, mixed lymphocyte reaction, IL-12 secretion function, to SD rat bone marrow mesenchymal stem cells to DC differentiation as a control. Results: Spermatozoa and glitches appeared in the induced EMSCs and became semi-adherent. SEM showed long cells protruding and many glitches; their surface markers were similar to bone marrow-derived DCs; mixed lymphocyte reaction proved to stimulate Lymphocyte proliferation; induced EMSC have IL-12 secretion function. Conclusion: EMSCs can induce DC differentiation in vitro.
